چکیده
Pulmonary tuberculosis (TB) is a chronic respiratory infectious disease. Certain microRNAs (miRNAs or miRs) are reported to be involved in regulating TB progression. The present study aimed evaluate the diagnostic potential of miR?125b pulmonary TB. expression levels and Raf1 proto?oncogene serine/threonine protein kinase (RAF1) were analyzed via reverse transcription?quantitative (RT?q)PCR patients with correlation between clinical indicators was investigated, receiver operating characteristic (ROC) curve used value miR?125b. relationship RAF1 examined using dual luciferase reporter gene assay. IL?6, TNF??, NF??B IFN?? detected ELISA kits, then NF??B, peripheral blood mononuclear cells (PBMCs) analyzed. Moreover, mRNA RT?qPCR western blotting. results demonstrated that decreased TB, while increased. Furthermore, associated sputum acid?fast bacillus smear. area under ROC 0.9413, sensitivity specificity for 90 92.5%, respectively. negatively correlated expression, inhibited by PBMCs. targeted regulate its levels. reversed role attenuating Thus, it suggested served important roles occurrence development decreasing inhibiting RAF1.

We investigated the diagnostic values of microRNA-31 in peripheral blood mononuclear cells (PBMCs) for pediatric pulmonary tuberculosis in Chinese patients. Sixty-five children with TB were selected for this study, which was conducted at the Department of Infectious Diseases People's Hospital of Laiwu City between December 2013 and December 2014. Sixty healthy children, selected in parallel, se...

Cryopreservation is the method of choice for long term storage of human PBMCs. This study was designed to compare the different combinations of variables affecting the cryopreservation of PBMCs samples. The viability of PBMCs separated from 2×5 ml peripheral blood samples obtained from 16 healthy adult volunteers, were measured using trypan blue dye exclusion method just before freezing with ...
متن کاملDiagnostic Value of the lncRNA NEAT1 in Peripheral Blood Mononuclear Cells of Patients with Sepsis
Background This study aims to evaluate the diagnostic value of nuclear-enriched abundant transcript 1 (NEAT1) expression in peripheral blood mononuclear cells (PBMCs) for the early diagnosis of sepsis. Methods A total of 59 patients with sepsis, 52 noninfectious SIRS patients, and 56 healthy controls were recruited fort this study. The levels of NEAT1 expression in PBMCs were measured using q...
متن کاملConditioned Medium from Cultured Colorectal Cancer Cells Affects Peripheral Blood Mononuclear Cells Inflammatory Phenotype in Vitro
Background: Colorectal cancer (CRC) is the third most common cancer worldwide. Studies have indicated that immune cells and soluble factors play a key role in maintaining the balance between tumor-promoting inflammation and anti-tumor immunity. It has been shown that secreted cytokines from CRC cell lines could affect peripheral blood mononuclear cells (PBMCs), monocytes, and macrophages phenot...
متن کاملPeripheral Blood Mononuclear Cells
Numerous cell types are involved in maintenance of the intestinal tissue. However, the main players are cells of the epithelial lining and the immune system. Human peripheral blood mononuclear cells (PBMCs) are used to investigate the effect of food bioactives on various immune cells. These cells are easily isolated from blood of healthy donors or buffy coats (leukocyte concentrates, a by-produ...
